Optical pumping of He-3 produces large (hyper) nuclear-spin polarizations independent of the magnetic resonance imaging (MRI) field strength. This allows lung MRI to be performed at reduced fields with many associated benefits, such as lower tissue susceptibility gradients and decreased power absorption rates. Here we present results of 2D imaging as well as accurate 1D gas diffusion mapping of the human lung using He-3 at very low field (3 mT). Furthermore, measurements of transverse relaxation in zero applied gradient are shown to accurately track pulmonary oxygen partial pressure, opening the way for novel imaging sequences.
We present NMR measurements of the diffusion of hyperpolarized helium-3 in the human lung performed at fields much lower than those of conventional MRI scanners. The measurements were made on standing subjects using homebuilt apparatus operating at 3 mT. Oxygen-limited transverse relaxation (T2 up to 15-35 s) could be measured in-vivo. Accurate global diffusion measurements have been performed in-vivo and in-vitro. 1D ADC mapping with high SNR demonstrates the real possibility of performing quality lung imaging at extremely low fields.
Purpose: Acquisition timing and B$_1$ calibration are two key factors that affect the quality and accuracy of hyperpolarized $^{13}$C MRI. The goal of this project was to develop a new approach using regional bolus tracking to trigger Bloch-Siegert B$_1$ mapping and real-time B$_1$ calibration based on regional B$_1$ measurements, followed by dynamic imaging of hyperpolarized $^{13}C$ metabolites in vivo. Methods: The proposed approach was implemented on a system which allows real-time data processing and real-time control on the sequence. Real-time center frequency calibration upon the bolus …
It was demonstrated that nonpersistent radicals can be generated in frozen solutions of metabolites such as pyruvate by irradiation with ultraviolet (UV) light, enabling radical-free dissolution DNP. Although pyruvate is endogenous, an excess of additional pyruvate may perturb metabolic processes, making it potentially unsuitable as a polarizing agent when studying fatty acids or carbohydrate metabolism. Therefore, the aim of the study was to characterize solutions containing endogenously-occurring alternatives to pyruvate as UV-induced nonpersistent radical precursors for in vivo hyperpolariz…
The human lung and its functions are extremely sensitive to orientation and posture, and debate continues as to the role of gravity and the surrounding anatomy in determining lung function and heterogeneity of perfusion and ventilation. However, study of these effects is difficult. The conventional high-field magnets used for most hyperpolarized 3He MRI of the human lung, and most other common radiological imaging modalities including PET and CT, restrict subjects to lying horizontally, minimizing most gravitational effects. In this paper, we briefly review the motivation for posture-dependent…
Nanodiamond is poised to become an attractive material for hyperpolarized 13C MRI if large nuclear polarizations can be achieved without the accompanying rapid spin-relaxation driven by paramagnetic species. Here we report enhanced and long-lived 13C polarization in synthetic nanodiamonds tailored by acid-cleaning and air-oxidation protocols. Our results separate the contributions of different paramagnetic species on the polarization behavior, identifying the importance of substitutional nitrogen defect centers in the nanodiamond core. These results are likely of use in the development of nano…
Through inhalation of, e.g., hyperpolarized $^3$He, it is possible to acquire gas diffusion magnetic resonance measurements that depend on the local geometry in the vast network of microscopic airways that form the respiratory zone of the human lung. Here, we demonstrate that this can be used to determine the dimensions (length and radius) of these airways noninvasively. Specifically, the above technique allows measurement of the weighted time-dependent diffusion coefficient (also called the apparent diffusion coefficient), which we here derive in analytic form using symmetries in the airway n…
Surface-functionalized nanomaterials can act as theranostic agents that detect disease and track biological processes using hyperpolarized magnetic resonance imaging (MRI). Candidate materials are sparse however, requiring spinful nuclei with long spin-lattice relaxation (T1) and spin-dephasing times (T2), together with a reservoir of electrons to impart hyperpolarization. Here, we demonstrate the versatility of the nanodiamond material system for hyperpolarized 13C MRI, making use of its intrinsic paramagnetic defect centers, hours-long nuclear T1 times, and T2 times suitable for spatially re…
Purpose: To develop an approach for improving the resolution and sensitivity of hyperpolarized 13C MRSI based on a priori anatomical information derived from featured, water-based 1H images. Methods: A reconstruction algorithm exploiting 1H MRI for the redefinition of the 13C MRSI anatomies was developed, based on a modification of the Spectroscopy with Linear Algebraic Modeling (SLAM) principle. To enhance 13C spatial resolution and reduce spillover effects without compromising SNR, this model was extended by endowing it with a search allowing smooth variations in the 13C MR intensity within …
The main findings of the european PHIL project (Polarised Helium to Image the Lung) are reported. State of the art optical pumping techniques for polarising ^3He gas are described. MRI methodological improvements allow dynamical ventilation images with a good resolution, ultimately limited by gas diffusion. Diffusion imaging appears as a robust method of lung diagnosis. A discussion of the potential advantage of low field MRI is presented. Selected PHIL results for emphysema are given, with the perspectives that this joint work opens up for the future of respiratory medicine.
Silicon-based micro and nanoparticles have gained popularity in a wide range of biomedical applications due to their biocompatibility and biodegradability in-vivo, as well as a flexible surface chemistry, which allows drug loading, functionalization and targeting. Here we report direct in-vivo imaging of hyperpolarized 29Si nuclei in silicon microparticles by MRI. Natural physical properties of silicon provide surface electronic states for dynamic nuclear polarization (DNP), extremely long depolarization times, insensitivity to the in-vivo environment or particle tumbling, and surfaces favorab…
The human lung and its functions are extremely sensitive to gravity, however the conventional high-field magnets used for most laser-polarized 3He MRI of the human lung restrict subjects to lying horizontally. Imaging of human lungs using inhaled laser-polarized 3He gas is demonstrated in an open-access very-low-magnetic-field (< 5 mT) MRI instrument. This prototype device employs a simple, low-cost electromagnet, with an open geometry that allows variation of the orientation of the imaging subject in a two-dimensional plane. As a demonstration, two-dimensional lung images were acquired with 4…

After being hyperpolarized using the technique of Metastability Exchange Optical Pumping (MEOP), 3He can be used as a contrast agent for lung magnetic resonance imaging (MRI). MEOP is usually performed at low magnetic field (~ 1 mT) and low pressure (~ 1 mbar), which results in a low magnetization production rate. A delicate polarization-preserving step of compression is also required. It was demonstrated in sealed cells that high nuclear polarization values can be obtained at higher pressures with MEOP, if performed at high magnetic field (non-standard conditions). In this work the feasibilit…

Magnetic resonance imaging at microtesla fields is a promising imaging method that combines the pre-polarization technique and broadband signal reception by SQUID sensors to enable in vivo MRI at microtesla-range magnetic fields. Despite significant advances, the potential of microtesla MRI for biomedical imaging is limited by its insufficient signal-to-noise ratio due to a relatively low sample polarization. Dynamic nuclear polarization (DNP) is a widely used approach that allows polarization enhancement by two-four orders of magnitude without an increase in the polarizing field strength. In …

Diffusion MRI measurements using hyperpolarized gases are generally acquired during patient breath hold, which yields a compromise between achievable image resolution, lung coverage and number of b-values. In this work, we propose a novel method that accelerates the acquisition of MR diffusion data by undersampling in both spatial and b-value dimensions, thanks to incorporating knowledge about the signal decay into the reconstruction (SIDER). SIDER is compared to total variation (TV) reconstruction by assessing their effect on both the recovery of ventilation images and estimated mean alveolar…
Purpose: Functional imaging is emerging as an important tool for lung cancer treatment planning and evaluation. Compared with traditional methods such as nuclear medicine ventilation-perfusion (VQ), positron emission tomography (PET), single photon emission computer tomography (SPECT), or magnetic resonance imaging (MRI), which use contrast agents to form 2D or 3D functional images, ventilation imaging obtained from 4DCT lung images is convenient and cost-effective because of its availability during radiation treatment planning. Current methods of obtaining ventilation images from 4DCT lung im…
