Inferring causal relationships or related associations from observational data can be invalidated by the existence of hidden confounding. We focus on a high-dimensional linear regression setting, where the measured covariates are affected by hidden confounding and propose the {\em Doubly Debiased Lasso} estimator for individual components of the regression coefficient vector. Our advocated method simultaneously corrects both the bias due to estimation of high-dimensional parameters as well as the bias caused by the hidden confounding. We establish its asymptotic normality and also prove that it is efficient in the Gauss-Markov sense. The validity of our methodology relies on a dense confounding assumption, i.e. that every confounding variable affects many covariates. The finite sample performance is illustrated with an extensive simulation study and a genomic application.
We consider statistical inference for a single coordinate of regression coefficients in high-dimensional linear models. Recently, the debiased estimators are popularly used for constructing confidence intervals and hypothesis testing in high-dimensional models. However, some representative numerical experiments show that they tend to be biased for large coefficients, especially when the number of large coefficients dominates the number of small coefficients. In this paper, we propose a modified debiased Lasso estimator based on bootstrap. Let us denote the proposed estimator BS-DB for short. W…

We consider the problem of variable selection in high-dimensional statistical models where the goal is to report a set of variables, out of many predictors $X_1, \dotsc, X_p$, that are relevant to a response of interest. For linear high-dimensional model, where the number of parameters exceeds the number of samples $(p>n)$, we propose a procedure for variables selection and prove that it controls the "directional" false discovery rate (FDR) below a pre-assigned significance level $q\in [0,1]$. We further analyze the statistical power of our framework and show that for designs with subgaussian …
We consider the problem of estimating a low-dimensional parameter in high-dimensional linear regression. Constructing an approximately unbiased estimate of the parameter of interest is a crucial step towards performing statistical inference. Several authors suggest to orthogonalize both the variable of interest and the outcome with respect to the nuisance variables, and then regress the residual outcome with respect to the residual variable. This is possible if the covariance structure of the regressors is perfectly known, or is sufficiently structured that it can be estimated accurately from …
We study the finite sample behavior of Lasso-based inference methods such as post double Lasso and debiased Lasso. We show that these methods can exhibit substantial omitted variable biases (OVBs) due to Lasso not selecting relevant controls. This phenomenon can occur even when the coefficients are sparse and the sample size is large and larger than the number of controls. Therefore, relying on the existing asymptotic inference theory can be problematic in empirical applications. We compare the Lasso-based inference methods to modern high-dimensional OLS-based methods and provide practical gui…
We consider the high-dimensional linear regression model $Y = X β^0 + ε$ with Gaussian noise $ε$ and Gaussian random design $X$. We assume that $Σ:= E X^T X / n$ is non-singular and write its inverse as $Θ:= Σ^{-1}$. The parameter of interest is the first component $β_1^0$ of $β^0$. We show that in the high-dimensional case the asymptotic variance of a debiased Lasso estimator can be smaller than $Θ_{1,1}$. For some special such cases we establish asymptotic efficiency. The conditions include $β^0$ being sparse and the first column $Θ_1$ of $Θ$ being not sparse. These conditions depend on whet…
We consider inference about coefficients on a small number of variables of interest in a linear panel data model with additive unobserved individual and time specific effects and a large number of additional time-varying confounding variables. We allow the number of these additional confounding variables to be larger than the sample size, and suppose that, in addition to unrestricted time and individual specific effects, these confounding variables are generated by a small number of common factors and high-dimensional weakly-dependent disturbances. We allow that both the factors and the distur…
This paper deals with prediction of anopheles number using environmental and climate variables. The variables selection is performed by an automatic machine learning method based on Lasso and stratified two levels cross validation. Selected variables are debiased while the predictionis generated by simple GLM (Generalized linear model). Finally, the results reveal to be qualitatively better, at selection, the prediction, and the CPU time point of view than those obtained by B-GLM method.

While linear mixed model (LMM) has shown a competitive performance in correcting spurious associations raised by population stratification, family structures, and cryptic relatedness, more challenges are still to be addressed regarding the complex structure of genotypic and phenotypic data. For example, geneticists have discovered that some clusters of phenotypes are more co-expressed than others. Hence, a joint analysis that can utilize such relatedness information in a heterogeneous data set is crucial for genetic modeling. We proposed the sparse graph-structured linear mixed model (sGLMM) t…
Performing statistical inference in high-dimension is an outstanding challenge. A major source of difficulty is the absence of precise information on the distribution of high-dimensional estimators. Here, we consider linear regression in the high-dimensional regime $p\gg n$. In this context, we would like to perform inference on a high-dimensional parameters vector $θ^*\in{\mathbb R}^p$. Important progress has been achieved in computing confidence intervals for single coordinates $θ^*_i$. A key role in these new methods is played by a certain debiased estimator $\hatθ^{\rm d}$ that is construc…
We propose two semiparametric versions of the debiased Lasso procedure for the model $Y_i = X_iβ_0 + g_0(Z_i) + ε_i$, where $β_0$ is high dimensional but sparse (exactly or approximately). Both versions are shown to have the same asymptotic normal distribution and do not require the minimal signal condition for statistical inference of any component in $β_0$. Our method also works when $Z_i$ is high dimensional provided that the function classes $E(X_{ij} |Z_i)$s and $E(Y_i|Z_i)$ belong to exhibit certain sparsity features, e.g., a sparse additive decomposition structure. We further develop a …
We consider random sample splitting for estimation and inference in high dimensional generalized linear models, where we first apply the lasso to select a submodel using one subsample and then apply the debiased lasso to fit the selected model using the remaining subsample. We show that, no matter including a prespecified subset of regression coefficients or not, the debiased lasso estimation of the selected submodel after a single splitting follows a normal distribution asymptotically. Furthermore, for a set of prespecified regression coefficients, we show that a multiple splitting procedure …
Exploring the genetic basis of heritable traits remains one of the central challenges in biomedical research. In simple cases, single polymorphic loci explain a significant fraction of the phenotype variability. However, many traits of interest appear to be subject to multifactorial control by groups of genetic loci instead. Accurate detection of such multivariate associations is nontrivial and often hindered by limited power. At the same time, confounding influences such as population structure cause spurious association signals that result in false positive findings if they are not accounted…
Zero-inflated explanatory variables are common in fields such as ecology and finance. In this paper we address the problem of having excess of zero values in some explanatory variables which are subject to multioutcome lasso-regularized variable selection. Briefly, the problem results from the failure of the lasso-type of shrinkage methods to recognize any difference between zero value occurring either in the regression coefficient or in the corresponding value of the explanatory variable. This kind of confounding will obviously increase number of false positives - all non-zero regression coef…
It is of particular interests in many application fields to draw doubly robust inference of a logistic partially linear model with the predictor specified as combination of a targeted low dimensional linear parametric function and a nuisance nonparametric function. In recent, Tan (2019) proposed a simple and flexible doubly robust estimator for this purpose. They introduced the two nuisance models, i.e. nonparametric component in the logistic model and conditional mean of the exposure covariates given the other covariates and fixed response, and specified them as fixed dimensional parametric m…
We devise a one-shot approach to distributed sparse regression in the high-dimensional setting. The key idea is to average "debiased" or "desparsified" lasso estimators. We show the approach converges at the same rate as the lasso as long as the dataset is not split across too many machines. We also extend the approach to generalized linear models.
In this study, we propose an automatic learning method for variables selection based on Lasso in epidemiology context. One of the aim of this approach is to overcome the pretreatment of experts in medicine and epidemiology on collected data. These pretreatment consist in recoding some variables and to choose some interactions based on expertise. The approach proposed uses all available explanatory variables without treatment and generate automatically all interactions between them. This lead to high dimension. We use Lasso, one of the robust methods of variable selection in high dimension. To …
Marginal Structural Models for Electricity Demand under Treatment-Confounder Feedback: A Continuous-Treatment Outcome-Adaptive and Fused LASSO Approach
Standard high-dimensional regression methods assume that the underlying coefficient vector is sparse. This might not be true in some cases, in particular in presence of hidden, confounding variables. Such hidden confounding can be represented as a high-dimensional linear model where the sparse coefficient vector is perturbed. For this model, we develop and investigate a class of methods that are based on running the Lasso on preprocessed data. The preprocessing step consists of applying certain spectral transformations that change the singular values of the design matrix. We show that, under s…

The Scientific Registry of Transplant Recipients (SRTR) system has become a rich resource for understanding the complex mechanisms of graft failure after kidney transplant, a crucial step for allocating organs effectively and implementing appropriate care. As transplant centers that treated patients might strongly confound graft failures, Cox models stratified by centers can eliminate their confounding effects. Also, since recipient age is a proven non-modifiable risk factor, a common practice is to fit models separately by recipient age groups. The moderate sample sizes, relative to the numbe…
The Convex Gaussian Min-Max Theorem (CGMT) has emerged as a prominent theoretical tool for analyzing the precise stochastic behavior of various statistical estimators in the so-called high dimensional proportional regime, where the sample size and the signal dimension are of the same order. However, a well recognized limitation of the existing CGMT machinery rests in its stringent requirement on the exact Gaussianity of the design matrix, therefore rendering the obtained precise high dimensional asymptotics largely a specific Gaussian theory in various important statistical models. This paper …

Evidence-based decision making often relies on meta-analyzing multiple studies, which enables more precise estimation and investigation of generalizability. Integrative analysis of multiple heterogeneous studies is, however, highly challenging in the ultra high dimensional setting. The challenge is even more pronounced when the individual level data cannot be shared across studies, known as DataSHIELD constraint (Wolfson et al., 2010). Under sparse regression models that are assumed to be similar yet not identical across studies, we propose in this paper a novel integrative estimation procedur…
We study Granger causality testing for high-dimensional time series using regularized regressions. To perform proper inference, we rely on heteroskedasticity and autocorrelation consistent (HAC) estimation of the asymptotic variance and develop the inferential theory in the high-dimensional setting. To recognize the time series data structures we focus on the sparse-group LASSO estimator, which includes the LASSO and the group LASSO as special cases. We establish the debiased central limit theorem for low dimensional groups of regression coefficients and study the HAC estimator of the long-run…
