Coherent Anti-Stokes Raman Scattering (CARS) processes are ``coherent,'' but the phase of the anti-Stokes radiation is usually lost by most incoherent spectroscopic CARS measurements. We propose a novel Raman microscopy imaging method called Nonlinear Interferometric Vibrational Imaging, which measures Raman spectra by obtaining the temporal anti-Stokes signal through nonlinear interferometry. With a more complete knowledge of the anti-Stokes signal, we show through simulations that a high-resolution Raman spectrum can be obtained of a molecule in a single pulse using broadband radiation. This could be useful for identifying the three-dimensional spatial distribution of molecular species in tissue.
Interferometric out-of-focus imaging of ice particles is realized. The overlapping defocused images of two nearby ice particles are analyzed. If one particle is much smaller than the other one, the pattern constitutes an off-axis hologram whose analysis gives the exact size and shape of the biggest particle. Using two angles of view, we obtain a 3D description of the biggest ice particle.
Interferometric imaging is a well established method to image phase objects by mixing the image wavefront with a reference one on a CCD camera. It has also been applied to fast transient phenomena, mostly through the analysis of single interferograms. It is shown that for repetitive phenomena multiphase acquisition brings significant advantages. A 1 MHz focused sound field emitted by a hemispherical piezotransducer in water is imaged as an example. Quantitative image analysis provides high resolution sound field profiles. Pressure at focus determined by this method agrees with measurements fro…
Illumination coherence plays a major role in various imaging systems, from microscopy, metrology, digital holography, optical coherence tomography, to ultrasound imaging. Here, we present a systematic study on the effects of degrees of spatiotemporal coherence of an illumination (DSTCI) on imaging quality. An optical field with arbitrary DSTCI was decomposed into wavelets with constituent spatiotemporal frequencies, and the effects on image quality were quantitatively investigated. The results show the synergistic effects on reduction of speckle noise when DSTCI is decreased. This study presen…
Quantitative phase imaging (QPI) quantifies the sample-specific optical-phase-delay enabling objective studies of optically-transparent specimens such as biological samples, but lacks chemical sensitivity limiting its application to morphology-based diagnosis. We present wide-field molecular-vibrational microscopy realized in the framework of QPI utilizing mid-infrared photothermal effect. Our technique provides mid-infrared spectroscopic performance comparable to that of a conventional infrared spectrometer in the molecular fingerprint region of 1,450 - 1,600 cm-1 and realizes wide-field mole…
We describe a high-speed single-shot multi-frame interferometric imaging technique enabling multiple interferometric images with femtosecond exposure time over a 50 ns event window to be recorded following a single laser-induced excitation event. The stroboscopic illumination of a framing camera is made possible through the use of a doubling cavity which produces a femtosecond pulse train that is synchronized to the gated exposure windows of the individual frames of the camera. The imaging system utilizes a Michelson interferometer to extract phase and ultimately displacement information. We d…
An interferometric fluorescent microscope and a novel theoretic image reconstruction approach were developed and used to obtain super-resolution images of live biological samples and to enable dynamic real time tracking. The tracking utilizes the information stored in the interference pattern of both the illuminating incoherent light and the emitted light. By periodically shifting the interferometer phase and a phase retrieval algorithm we obtain information that allow localization with sub-2 nm axial resolution at 5 Hz.
Energy carrier evolution is crucial for material performance. Ultrafast microscopy has been widely applied to visualize the spatiotemporal evolution of energy carriers. However, direct imaging of small amounts of energy carriers on nanoscale remains difficult due to extremely weak transient signals. Here we present a method for ultrasensitive and high-throughput imaging of energy carrier evolution in space and time. This method combines femtosecond pump-probe techniques with interferometric scattering microscopy (iSCAT), named Femto-iSCAT. The interferometric principle and unique spatially-mod…
We report the imaging of tendon, a connective tissue rich in collagen type I proteins, with Interferometric Second Harmonic Generation (I-SHG) microscopy. We observed that the noncentrosymmetric structural organization can be maintained along the fibrillar axis over more than 150 μm, while in the transverse direction it is ~1-15 μm. Those results are explained by modeling tendon as a heterogeneous distribution of noncentrosymmetric nanocylinders (collagen fibrils) oriented along the fibrillar axis. The preservation of the noncentrosymmetric structural organization over multiple tens of microns…
Optical tomographic cross-sectional images of biological samples were made possible by interferometric imaging techniques such as Optical Coherence Tomography (OCT). Owing to its unprecedented view of the sample, OCT has become a gold standard, namely for human retinal imaging in the clinical environment. In this Letter, we present Optical Incoherence Tomography (OIT): a completely digital method extending the possibility to generate tomographic retinal cross-sections to non-interferometric imaging systems such as en-face AO-ophthalmoscopes. We demonstrate that OIT can be applied to different …
Our ability to optically interrogate nanoscopic objects is controlled by the difference between their extinction cross sections and the diffraction limited area to which light can be confined in the far field. We show that a partially transmissive spatial mask placed near the back focal plane of a high numerical aperture microscope objective enhances the extinction contrast of a scatterer near an interface by approximately $T^{-1/2}$, where T is the transmissivity of the mask. Numerical aperture based differentiation of background from scattered light represents a general approach to increasin…
We present a new technique for obtaining simultaneous multimodal quantitative phase and fluorescence microscopy of biological cells, providing both quantitative phase imaging and molecular specificity using a single camera. Our system is based on an interferometric multiplexing module, externally positioned at the exit of an optical microscope. In contrast to previous approaches, the presented technique allows conventional fluorescence imaging, rather than interferometric off-axis fluorescence imaging. We demonstrate the presented technique for imaging fluorescent beads and live biological cel…
Nonlinear optical methods, such as coherent anti-Stokes Raman scattering (CARS) and stimulated Raman scattering (SRS), are able to perform label free imaging, with chemical bonds specificity. Here, we demonstrate that the use of circularly polarized light allows to retrieve not only the chemical nature but also the symmetry of the probed sample, in a single shot measurement. Our symmetry-resolved scheme offers simple access to the local organization of vibrational bonds and as a result provides enhanced image contrast for anisotropic samples as well as an improved chemical selectivity. We quan…
High-dimensional biphoton states are promising resources for quantum applications, ranging from high-dimensional quantum communications to quantum imaging. A pivotal task is fully characterising these states, which is generally time-consuming and not scalable when projective measurement approaches are adopted. However, new advances in coincidence imaging technologies allow for overcoming these limitations by parallelising multiple measurements. Here, we introduce biphoton digital holography, in analogy to off-axis digital holography, where coincidence imaging of the superposition of an unknown…
Interferometric particle imaging is a widely used optical measuring technique for the sizing of poly-dispersed spherical particles like droplets and bubbles. In its conventional approach, the method is limited to forward-scattering angles and therefore, requiring a second optical access, restricting the range of possible applications. In the present work, this limitation of the scattering angle is addressed, showing that also other scattering angles, especially in the back-scatter region are applicable, expanding the technique to applications with only a single optical access. A general method…
This paper reported a general noninterferometric high-accuracy quantitative phase imaging (QPI) method for arbitrary complex valued objects. Given by a typical 4f optical configuration as the imaging system, three frames of small-window phase modulation are applied on the object Fourier spectrum so that redistributed intensity patterns are produced on the image plane, in which the object phase emerges at different degree. Then, an algebraic relationship that connects the object phase with the output intensity is established to provide us with an approximate closed form phase recovery. Further,…
Fourier domain mode-locking (FDML) has been a popular laser design for high speed optical frequency domain imaging (OFDI) but achieving long coherence lengths, and therefore imaging range, has been challenging. The narrow instantaneous linewidth of a frequency comb (FC) FDML laser could provide an attractive platform for high speed as well as long range OFDI. Unfortunately, aliasing artifacts arising from signals beyond the principle measurement depth of the free spectral range have prohibited the use of a FC FDML for imaging so far. To make the enhanced coherence length of FC FDML laser avail…
Interferometric scattering microscopy has been a very promising technology for highly sensitive label-free imaging of a broad spectrum of biological nanoparticles from proteins to viruses in a high-throughput manner. Although it can reveal the specimen's size and shape information, the chemical composition is inaccessible in interferometric measurements. Infrared spectroscopic imaging provides chemical specificity based on inherent chemical bond vibrations of specimens but lacks the ability to image and resolve individual nanoparticles due to long infrared wavelengths. Here, we describe a bond…
Currently, cellular action potentials are detected using either electrical recordings or exogenous fluorescent probes sensing calcium concentration or transmembrane voltage. Ca imaging has low temporal resolution, while voltage indicators are vulnerable to phototoxicity, photobleaching and heating. Here we report full-field interferometric imaging of individual action potentials by detecting the movement across the entire cell membrane. Using spike-triggered averaging of the movies synchronized to electrical recording, we demonstrate deformations of up to 3 nm (0.9 mrad) during the action pote…

The detection of spatial or temporal variations in very thin samples has important applications in the biological sciences. For example, cellular membranes exhibit changes in lipid composition and order, which in turn modulate their function in space and time. Simultaneous measurement of thickness and refractive index would be one way to observe these variations, yet doing it noninvasively remains an elusive goal. Here we present a microscopic-imaging technique to simultaneously measure the thickness and refractive index of thin layers in a spatially resolved manner using reflectometry. The he…
